چکیده

The ubiquitin-editing protein A20 limits the duration of NF-kB activation and is essential to control inflammatory responses. Recent experiments have established that A20 mediates its inhibitory function in a complex with two other proteins, TAX1BP1 and the E3 ligase Itch. In a paper published in this issue, it is shown that RNF11 is also a critical component of the A20 ubiquitin-editing complex. NF-kB transcription factors regulate the expression of numerous genes involved in inflammation and immunity (Li and Verma, 2002). Activation of NF-kB is normally transient, and persistent NF-kB activation is associated with several autoimmune diseases and cancer. The duration of NF-kB activation is controlled by a number of inhibitory proteins, including A20, which functions in a negative feedback loop to limit NF-kB activation. A20 is essential

The ubiquitin-editing enzyme A20 requires RNF11 to downregulate NF-kappaB signalling.

The RING domain protein RNF11 is overexpressed in breast cancers and promotes tumour growth factor-beta (TGF-beta) signalling. RNF11 has been proposed to regulate TGF-beta signalling by interacting with HECT- and SCF-type E3 ligases; however, the role of RNF11 in other signalling pathways is poorly understood. Here, we demonstrate a novel function of RNF11 as a negative regulator of NF-kappaB a...

Expression of the nuclear factor-κB inhibitor A20 is altered in the cystic fibrosis epithelium.

A20 is a lipopolysaccharide (LPS)-inducible, cytoplasmic zinc finger protein, which inhibits Toll-like receptor-activated nuclear factor (NF)-κB signalling by deubiquitinating tumour necrosis factor receptor-associated factor (TRAF)-6. The action of A20 is facilitated by complex formation with ring finger protein (RNF)-11, Itch and TAX-1 binding protein-1 (TAX1BP1). This study investigated whet...

Ring Finger Protein 11 Inhibits Melanocortin 3 and 4 Receptor Signaling

Intact melanocortin signaling via the G protein-coupled receptors (GPCRs), melanocortin receptor 4 (MC4R), and melanocortin receptor 3 (MC3R) is crucial for body weight maintenance. So far, no connection between melanocortin signaling and hypothalamic inflammation has been reported. Using a bimolecular fluorescence complementation library screen, we identified a new interaction partner for thes...

Involvement of Ubiquitin-Editing Protein A20 in Modulating Inflammation in Rat Cochlea Associated with Silver Nanoparticle-Induced CD68 Upregulation and TLR4 Activation

Silver nanoparticles (AgNPs) were shown to temporarily impair the biological barriers in the skin of the external ear canal, mucosa of the middle ear, and inner ear, causing partially reversible hearing loss after delivery into the middle ear. The current study aimed to elucidate the molecular mechanism, emphasizing the TLR signaling pathways in association with the potential recruitment of mac...

Financial Incentives: Only One Piece of the Workplace Wellness Puzzle; Comment on “Corporate Wellness Programs: Implementation Challenges in the Modern American Workplace”

In this commentary, we argue that financial incentives are only one of many key components that employers should consider when designing and implementing a workplace wellness program. Strategies such as social encouragement and providing token rewards may also be effective in improving awareness and engagement. Should employers choose to utilize financial incentives, they should tailor them to ...
